I'm listing a digital piano available for pickup only. Starting auction price would be $450. Is it possible to request a $100 deposit and the balance when they pick up?  I've never sold an item over $200 before and I'm not sure how people feel about paying this much outright for an item.

 

Has anyone had experience with this?

No, it's not possible to require a deposit on a regular item on eBay. You will ask for payment on pickup, in cash.

I'm not sure how people feel about paying a deposit on something they don't have...

 

As advised, you do not want to take a PayPal payment for a pick-up item as you would have no Seller Protection. You want cash upon pickup...in full.
